Studies on Diuretic Action of Rutin and Quercetin-pentacetate



Abstract
Rutin and its derivatives have been used for treatment of inflammation, diabetes mellitus, allergy, headache, virus infection, chemical oncogenesis, X-ray irradiation, stomach ulcer, duodental ulcer etc. The chemical constituents so far on Sophora flos are known in isolation of flavonoids rutin (quercetin 3-rutinoside) and triterpenoids, betulin, betulinic acid, sophoradiol. Preparations which contain flavonoids as the principal physiologically active constituents have been used by physician in attempts to treat humen diseases. Some pharmacological works have also been done, however, few papers on its cardio-diuretic effects have been reported.
Therefore, we intented to determine the pharmacological action of rutin and quercetinpentacetate, especially, their diuretic effects in rabbits. Quercetin-pentacetate synthesized through acetylation and hydrolysis process from the start of a rutin component. Two components were intraveously injected to the rabbit in the dose of 20 mg/kg, 10 mg/kg and 5 mg/kg. The variations of urine volume and minerals (Na+, K+, and Cl-) in urine were measured at an interval of 15 minutes for 4 hours after the treatment of two components.
The observed results are as follows:
(1) The experimental group treated rutin in urine volume as well as excretion rate of urin-
ary minerals (Nar, K+ and Cl-) demonstrated diuretic action of relatively long duration.
(2) In a dose of 20mg/kg, significant variations of urine volume and (Nat, K+ and Cl-)
were observed in the quercetin-pentacetate treated group, compared with those of rutin
treated group throughout the experiments. But, in a dose of 10mg/kg and 5mg/kg, the
slight increment on diuretic action of rabbit was observed.
In conclusion, our present data suggest that diuretic action in a case of substitution acetyl ester for the alcohol group in positions 3, 5, 7, 3¢¥ and 4¢¥ of rutin is increased with the administration of quercetin-pentacetate (20mg/kg).
